New reactions for the synthesis of polycyclic indole alkaloids

Polycyclic indole alkaloids represent a diverse range of biologically important natural products. Herein we present the development of simple synthons that facilitate rapid entry into the Aspidosperma alkaloid framework. Our studies have focused on Lewis acid activation of N,O-trimethylsilyl acetals derived from tryptamine and simple carboxylic acids. The scope and limitations of the resulting cascade reactions will be discussed.
