Structural study of ABH proteins by utilizing chemical crosslinking strategy

Two human homologs of the AlkB protein, ABH2 and ABH3, are known to bind and repair alkylated base damage on dsDNA, ssDNA, and ssRNA. However it's not clear how these proteins interact with different nucleic acid structures and how they search for the damaged base in both dsDNA and ssDNA. We have developed and applied a chemical cross-linking technique to study the interaction of these proteins with DNA and the structures of their DNA complexes.
