Total synthesis of the hexacyclinols and their role in advancing endoperoxide antimalarials

Endoperoxide-containing terpenes including artemisinin and yinghaosu A currently offer a first level of defense against plasmodial infections. While the development of synthetic analogs has suggests cost effective remedies for the treatment of Malaria, the instability of the functional endoperoxides within these materials has complicated their use in the field. Within this lecture, a mechanistic approach to malarial-regulation is presented through the total synthesis of a novel class of endoperoxide containing terpenes, the hexacyclinols.
