Spirastrellolide A, a novel macrolide isolated from the marine sponge Spirastrella coccinea, has been demonstrated to be a potent and selective inhibitor of protein phosphatase 2A (IC50 = 1nM). Key intermediates are envisioned to be synthesized by implementing methodology previously discovered in our laboratory. Our convergent design provides an efficient and flexible approach that will enable a rapid assembly of the C1-C15 fragment, and allow easy access to the desired diastereomers.
