Studies toward the total synthesis and absolute configurational determination of (+)-varitriol

First isolated from the marine derived strain of the fungus Emericella variecolor in 2002 by Barrero, varitriol (1) represents a potent anti-tumor agent which exhibited significant cytotoxic activity (mg/ml) against RXF 393 (renal cancer), T-47D (breast cancer), and SNB-75 (CNS cancer) human carcinoma cell lines. In addition to the impressive levels of biological activity, the highly unique and somewhat simple structure of 1 makes it an attractive target for total synthesis. Towards this end, synthetic strategies aimed at the total synthesis of 1 will be discussed in greater detail.
