Synthesis of C-6 heteroaryl nucleosides

Nitrogen-containing heterocycles are subunits found in numerous natural products and in many biologically active pharmaceuticals. Some heteroaryl purine nucleosides are synthetically useful substrates for nucleophilic aromatic substitution and metal catalyzed reactions leading to other modified nucleosides. However, various heteroaryl nucleosides have been prepared by widely different synthetic procedures and to date there is no single unifying method for the synthesis of a broad range of C-6 heteroaryl nucleosides. This work will demonstrate a simple, palladium-based C-N bond formation method that can be utilized for the synthesis of structurally diverse C-6 heterocyclic nucleosides.
