| We report here efficient fluorous tagging reagents that facilitate purification of peptides synthesized on the solid phase. The reagents are based on perfluoroalkylmethylene iodonium salts, which are very stable in storage and as solutions, yet potent alkylating agents under reaction conditions. While retaining standard solvents and equipment, we show potent capping by fluorous iodonium salts, which render the capped products significantly different from the desired full length products. Such capping reagents offer significant advantages in purification and tagging of peptides. Several practical examples where the SPPS is markedly improved and purification simplified will be discussed and insights into the mechanism of capping and reactivity detailed. |
