Synthetic studies toward the antitumor antibiotic FR901464

FR901464 is a potent antitumor antibiotic natural product. It induces apoptosis in a variety of tumor cell lines at 1 nM concentrations. However, the unique biological mechanism of FR901464 is unknown. We are currently synthesizing FR901464 in a convergent manner utilizing chiral catalysis as well the chiral pool. Details of the synthetic progress and biological studies of the target will be described. A highly diastereoselective method using NBS to form tetrahydropyranyl derivatives from alkenyl ketones will also be described.
