Design and synthesis of antimalarial agents based on Manzamine A

Manzamine A is a novel marine alkaloid that exhibits potent antimalarial activity (IC50 = 0.56 ng/ml against P. falciparum). Since its isolation in 1986, manzamine A has attracted a great deal of interest in the synthetic community. In 1998, our laboratory completed the first total synthesis of manzamine A based on an intramolecular vinylogous amide photocycloaddition-fragmentation-Mannich closure sequence. Our current work focuses on determining the structure-activity relationship of various partial structures of manzamine A against the malaria causing pathogen P. falciparum. We have designed and synthesized five manzamine A analogues of varying complexity. The biological activity of four of these analogues will be presented.
