Azaspiracid-1, first observed in the mid-1990’s off the coast of western Ireland, has garnered considerable biological and synthetic interest due to its unique structural architecture and significant toxicity profile. Further interest in this area has been fueled by the mis-assignment of the natural product. The successful construction of the transoidal bisspiroketal of the ABCD ring system will be detailed. In addition, the synthesis of the ABCDE ring system will be reported and progress toward the determination of the exact structure of azaspiracid-1 will be discussed.
