Novel conformationally constrained pseudopeptides derivatives of the tripeptide pYVN were synthesized as antagonists for the Grb2-SH2 domain. Extended and reverse turn peptide conformations were mimicked by trans- and cis- cyclopropanes, respectively. The pseudopeptides were evaluated using isothermal microcalorimetry and X-ray crystallography.
