Toward the synthesis of nucleoside containing a metal chelator

Despite the advances in structural analysis of RNA and RNA-complexes with proteins and small molecules over the last few years, there still exists a need for time-effective spectroscopic methods for RNA structural studies. We are interested in site-specific incorporation of metal ions into RNA as spectroscopic probes, which could find applications in the study of the structure of RNA and its complexes with protein and small molecules. We will describe the preparation of a nucleoside containing an analog of the metal chelator ethylene diamine tetracarboxylate (EDTA). The EDTA moiety binds metal ions with high affinity and this nucleoside will be used to bind a metal ion at a specific site in the nucleic acid.
