Carbonic Anhydrase (CA) is a ubiquitous zinc enzyme that catalyzes the reversible hydration of CO2. Considerable effort has been given to identifying metal complexes that mimic the active site of CA. In this regard, macrocycles with multi-Lewis basic sites have been widely used, as they are more preorganized than their acyclic analogues. As part of our work toward the mimicry of Carbonic Anhydrase, we have designed a class of macrocyclic ligands that contain three pyridine rings for coordination, and a chiral center to engender a chiral environment at the metal binding site. The synthesis of these ligands, as well as their Zn2+ binding studies, will be presented.![]() |
