Parallel solution-phase synthesis of tetra-substituted purines

Purine ring containing compounds are important structural units in bio-molecules showing wide-range of pharmacological activities. They have been studied extensively in the literature. However, few synthetic methods were reported for fully substituted purine analog so far. This report presented a convenient preparation of tetra-substituted purines in solution phase as shown in the following scheme starting from a series of 2-substituted 5-amino-4,6-dichloropyrimidines. Mono-substitution by an amine followed by cyclization to form the purine ring and a final amine substitution yielded tetra-substituted purine libraries.
