High-throughput purification of a library by LC-MS systems using a double screening method

Because of the large difference in property for each individual compound within a library, it is sometimes hard to get the whole library purified by HPLC using same gradient profile. We have developed a pH screening method that helps effectively resolve each mixture within a small library in a high throughput fashion. A small pyrazole library of 16 individual mixture has been successfully purified using such a method. A parallel post-treatment may further remove any reagent residue that may interfere in the HTS process of the library to ensure high quality and avoid false results.
