The advantages of solid state synthesis over the solution phase are: simple purification of the end product by filtration; lower reaction time; possibility for automation; and development of a large compound library for drug discovery. Solid-phase synthesis of peptides, nucleotides and libraries of organic compounds are well documented. On the other hand, lipid synthesis employing this methodology has not been explored in detail. The solid phase synthesis was used for the preparation of a variety of lipids, will be presented. Some of these lipids were made previously in a solution phase. Using the solid support synthesis of these lipids, it was established that the reaction time in most of the steps was reduced a least by half, while the overall yield of the end-product was elevated. In addition, this method reduces purification steps compared to the solution method. ![]() |
