Apoptolidin was isolated in 1997 from Nocardiopsis sp. and was determined to induce apoptotic cell death in rat glia cells transformed with the E1A oncogene (IC50=11ng/ml). More recently, Kholsa and co-workers identified the mitochondrial F1F0 ATPase as a target to explain its unique biological function. Our research group is interested in developing a novel synthetic route to the apoptolidin aglycon in efforts to study its biological and chemical characteristics. Recent progress towards this end will be presented.
